PT-141 (Bremelanotide) is a synthetic peptide analog of alpha-MSH (melanocyte-stimulating hormone). PT-141 is unique among sexual health compounds in that it does not act on vascular smooth muscle (like PDE-5 inhibitors) but instead activates central melanocortin pathways. The MC4 receptor is involved in reward and motivation circuits, and PT-141 appears to modulate these pathways to enhance sexual desire and arousal. The peptide has demonstrated efficacy in both male and female subjects in clinical research. It was approved by the FDA in 2019 for HSDD in women (as Vyleesi).
